GLP-1 10 mg for receptor pharmacology
The GLP-1 receptor couples mainly to Gs, so agonist binding raises intracellular cAMP. That makes cAMP accumulation, measured by HTRF, luminescence or reporter-gene assays, the standard functional readout, with receptor internalisation and beta-arrestin recruitment as common secondary measures. A well-characterised agonist is the reference point every one of those assays needs.
Class B receptors bind their peptide ligands in two steps, first through a large extracellular domain and then within the transmembrane core. Differences in how agonists engage those two sites are behind much of the current work on biased signalling at this receptor, and a consistent supply of agonist is what lets those comparisons hold across months of experiments.
